Usage

Tafinlar (Dabrafenib Mesylate) is a prescription medication used to treat various cancers, including skin, thyroid, lung, solid tumors, and brain cancer. It is also used in combination with Mekinist to treat skin cancer called melanoma that has metastasized to other parts of the body and to prevent melanoma that has a certain type of abnormal "BRAF" gene from coming back after the cancer has been removed by surgery. The active ingredient in Tafinlar is Dabrafenib Mesylate. Dabrafenib Mesylate belongs to the class of medication called antineoplastic (BRAF Kinase Inhibitor). It works by blocking the action of an abnormal protein that signals cancer cells to multiply, thereby stopping the spread of cancer cells.

Side Effects and Precautions

The common side effects of Tafinlar are nausea, vomiting, headache, alopecia, cough, rash, and backache.

Stop using Tafinlar and contact your doctor right away if you have any of the serious side effects such as:
• Loss of vision or visual disturbances;
• Shortness of breath, chest pain, or arm or leg swelling;
• Skin bump that bleeds or does not heal;
• Sudden changes in the appearance of your skin, including sores, lumps, warts, change in the size/color of a mole;
• Unintended weight gain

Warnings and Interactions

Before taking dabrafenib, tell your doctor or pharmacist if you are allergic to it or have any other allergies. Tafinlar comes in a capsule and should be taken orally with a cup of water. It is usually taken twice daily on an empty stomach, 1 hour before or 2 hours after a meal. Take your medication about 12 hours apart at around the exact times daily. Swallow the capsules whole; do not open, break, or crush them. Tell your doctor if you cannot swallow the tablets whole. Follow the directions on your prescription label carefully, and ask your doctor or pharmacist to explain any part you do not understand. Men and women of reproductive age should use a reliable form of non-hormonal birth control (such as condoms diaphragm with spermicide) during treatment and for 2 weeks after the last dose. Tafinlar may worsen your sugar level. It is advisable to check your blood sugar regularly and inform your doctor if there is a sudden rise in your blood sugar. Do not stop taking dabrafenib without talking to your doctor.

Before you start using Tafinlar, inform your doctor about your medical history, especially if you have:
• Bleeding problems;
• Diabetes mellitus;
• G6PD deficiency;
• Kidney problems;
• Liver problems

Tell your doctor about all your medications, including prescriptions, over-the-counter medications, herbal products, and vitamin supplements. Tell your doctor if you are pregnant or plan to become pregnant. Tafinlar should not be used during pregnancy. Tafinlar may harm an unborn baby. If you become pregnant, talk to your doctor right away about the risks and benefits of continuing this medication in pregnancy. It is unknown if Tafinlar passes into breast milk. Because of the possible risk to the infant, breastfeeding is not recommended while using this drug and for 2 weeks after the last dose. Consult your doctor before breastfeeding for alternative feeding practices. There may be a drug interaction between Tafinlar and other drugs such as tacrolimus, naloxegol, lonafarnib, fostemsavir, doravirine, praziquantel, and lefamulin.
