Usage

Cotellic (Cobimetinib Fumarate) is a prescription medication commonly used with Vemurafenib for the treatment of melanoma (a type of skin cancer) that has metastasis to other parts of the body or cannot be surgically removed. Cotellic is only given to patients whose melanoma is caused by the BRAF V600 mutation and must be combined with vemurafenib, a BRAF inhibitor. The active ingredient in Cotellic is Cobimetinib Fumarate. Cobimetinib Fumarate belongs to the group of medications called Protein Kinase inhibitors. It works by blocking the action of an abnormal protein that signals cancer cells to multiply. As a result, it slows down the growth and spread of the cancer.

Side Effects and Precautions

The common side effects of Cotellic are nausea, vomiting, changes in appetite, abdominal pain, dry skin, hair loss, and muscle pain.

Stop using Cotellic and contact your doctor right away if you have any of the severe side effects such as:
• Chronic cough, shortness of breath, or difficulty with breathing;
• Easy bruising;
• Symptoms of electrolytes derangement such as headache, confusion, severe constipation, slurred speech;
• Unexplained muscle pain;
• Worsening eyesight or partial visual loss;
• Worsening of your condition

Warnings and Interactions

Cotellic is a tablet that should be taken orally with or without food. You should know that Cotellic is given in a 28-day treatment cycle. Your doctor will determine how long to treat you with Cotellic. It is usually taken once daily for the first 21 days, followed by a 7-day break of a 28-day cycle. After that, take the Cotellic tablet at around the same time daily. Follow the directions on your prescription label carefully and ask your doctor to explain any part you do not understand. Do not take grapefruits or juice with Cotellic tablets. Do not use more than your prescribed dosage. Check your skin regularly while you are using Cotellic. Tell your doctor if you notice any new skin change, such as redness, sores that will not heal, or a mole that has changed in size or color. Continue to take this medication for the period it was prescribed, even if you feel better or symptoms remain the same. Do not miss your clinic appointment, and contact your doctor if you experience a severe side effect.

Before you start using Cotellic, inform your doctor about your medical history, especially if you have the following:
• Any chronic medical condition such as heart disease or abnormal heart rhythm;
• Bleeding disorders such as hemophilia or clotting problems;
• Eye problems such as cataracts or glaucoma;
• Immune system problems such as Crohn's disease, ulcerative colitis, or lupus;
• Kidney problems;
• Liver problems;
• Severe skin infections

Tell your doctor about all your medications, including prescriptions, over-the-counter medications, herbal products, and vitamin supplements. Tell your doctor if you are pregnant or plan to become pregnant. Cotellic can harm the unborn baby. Your doctor may do a pregnancy test before starting you on Cotellic medication. You should be on an approved contraceptive while taking Cotellic to avoid getting pregnant. Inform your doctor if you are breastfeeding or plan to breastfeed. It is not known if Cotellic passes into your breast milk. There may be a drug interaction between Cotellic and other drugs such as armodafinil, clarithromycin, darunavir, efavirenz, dexamethasone, rifampin, and tipranavir.
