Indications

Rapamune is a transplant medication used to prevent organ rejection in patients receiving kidney transplants.

Sirolimus is a mammalian target of rapamycin (mTOR) inhibitor that suppresses T-lymphocyte activity that occurs after antigenic and cytokine stimulation which stops the progression of the cell cycle and installs an immunosuppressive complex. These inhibitory actions allow rapamune to protect against organ rejection for those receiving kidneys and other allograft transplants by preventing your immune system from attacking new organs.

Uses and Dosage

The dose of Rapamune that is prescribed will vary depending on the individual being treated. Follow all your doctor’s instructions carefully to ensure the best results while taking sirolimus.

  • Swallow tablets whole
  • Can be taken with or without food but take Rapamune the same way each day
  • Adults with low to moderate immunologic risk tend to take a maintenance dose of 2mg daily
  • Maintenance doses should be preceded with a loading dose three times the amount, for example, 2mg once daily should be preceded by one dose of 6mg
  • Patients with high immunologic risk are recommended to take Rapamune with cyclosporine and corticosteroids for 12 months after transplantation
  • For combined therapy, patients should start by taking a loading dose of 15mg the day after transplantation, starting day two they should take 5mg once daily
  • Doses can be adjusted after 7 to 14 days of maintenance doses
  • When taking cyclosporine, wait at least 4 hours before taking your dose of Rapamune
  • Do not take more or less than prescribed by your doctor
  • Do not stop taking Rapamune or other anti-rejection medicines unless directed by your doctor
  • Store at room temperature away from children and pets

FAQ

How does Rapamune prevent rejection?

Rapamune helps prevent organ transplant rejection by inhibiting your body’s immune response and lowering your number of T-lymphocytes which attack new or foreign organs. Thanks to these inhibitory actions, the drug helps prevent rejection following kidney transplants.

What precautions should be taken when taking Rapamune?

You should avoid ingesting any grapefruit products during treatment with Rapamune because it can cause an increase in your blood concentration of sirolimus and lead to adverse reactions. This medication may also increase your risk of developing skin cancer, so it is important to avoid direct exposure to sunlight and UV rays during treatment.
